Arog’s lead molecule, crenolanib, is a type I TKI that selectively and potently inhibits signaling of wild-type and mutant isoforms of class III receptor tyrosine kinases (RTKs), FLT3 and PDGFRα/β. FLT3 is a class III RTK, and its signaling is considered important for the normal development of hematopoietic stem cells and progenitor cells. The FLT3 gene is one of the most frequently mutated genes (~30%) in AML. One such mutation, internal tandem duplications of FLT3 (FLT3-ITD), is a prognostic indicator associated with adverse disease outcome.
About PDGFRα D842V Mutations
PDGFRAD842V is a gain-of-function mutation that constitutively activates PDGFRA downstream signaling pathway. D842V is the most common PDGFRA mutation and is well-known to be resistant to imatinib and sunitinib.
